The stability of antibodies (Ab) diluted in buffers depends on a variety of factors.
The antibody diluent should :
• Minimise adhesion of the Ab to the wall of the tube
• Prevent proteolytic degradation of the Ab
• Increase microbial and chemical stability of the Ab preparation
• Ensure good spreading of the Ab solution on the tissue section
• For some tricky Ab, improve the IHC staining intensity
• Tackle the non specific binding of primary Ab on the tissue
• Inhibit growth of micro-organisms
A diluent buffer (Antibody Diluent B) has been specifically developed for certain primary antibodies like MLH-1, leading to significantly stronger signals in immunohistochemistry.
Antibodies diluted with Antibody Diluent B are primarily used in immunohistochemistry with formalin-fixed paraffin-embedded tissue sections, and also with frozen, HOPE-fixed, and cytological samples as well as in immunoblot procedures.

The Antibody Diluent fulfils all these needs
Immunohistochemical staining procedures often start with incubation of a blocking solution to reduce unspecific binding of the primary antibody to tissue sections. This step can be skipped if the primary antibody is diluted in Antibody Diluent.
The antibody diluent is stained with a green dye for a better identification on the slide.
Antibodies diluted in the Antibody Diluent (cat# ZUC025) are stable for several months. There is therefore no need to prepare working solutions every day or pipette very small amounts of concentrated antibodies.
