The Diagnosis of gastrointestinal stromal tumours (GIST) relies on clinical and morphological features as well as on the immunohistochemical detection of KIT(CD117) protein. Overexpression of this type III receptor tyrosine kinase caused by mutations in the corresponding gene leads to uncontrolled cell growth. Imatinib is a receptor tyrosine kinase inhibitor and a potent therapeutic agent against GIST.
However, in approximately 4 to 15 % of all clinical and morphological definite GIST the KIT protein is not detectable. Nevertheless some of these patients respond to therapy with Imatinib. Medeiros et al. show that the majority of GIST, which were tested negative for KIT, show mutations in the gene for the platelet-derived growth factor receptor-alpha (PDGFR-alpha).
West et al. and Espinosa et al. characterised DOG1, a protein with unknown function, as new sensitive and specific marker for GIST. Via labelling of DOG1 (Discovered On GIST-1) considerably more GIST cases could be detected than via labelling of KIT. GIST with both KIT and PDGFR-alpha mutation were detected. This means the number of false-negative GIST could be reduced by using DOG1 staining. More patients than before could profit from Imatinib therapy. DOG1 is easily labelled immunohistochemically with a polyclonal rabbit antibody on formalin-fixed tissue sections.

GPC3 is useful for differentiation of hepatocellular carcinomas (HCC) from non-malignant liver tissue and other primary and metastatic lesions. Recent publications describe that Glypican-3 is overexpressed in most HCC
whereas normal liver tissue and benign lesions such as dysplastic and cirrhotic nodules are negative for Glypican-3.

In most cases patient history as well as biochemical and histomorphological analyses provides enough data to carry out a diagnosis of parathyroid gland proliferations. But in some cases the immunohistochemical detection of Parathyroid Hormone (PTH) is very helpful, for example in tumours with unusual localisation, tumours with clear cell morphology or non-secretory tumours.
In the last months, some well-established antibodies for detection of PTH on FFPE were discontinued and no longer available on the market. To close this gap Zytomed Systems now offers a new monoclonal antibody of clone MRQ-31. Zytomed Systems new antibody against PTH ensures a reliable staining of Parathyroid Hormone on formalin-fixed paraffin-embedded tissue sections.

Uroplakins Ia, Ib, II and III are structural proteins of terminal differentiated urothelial cells. In normal urothelia they are expressed in the luminal cytoplasmic membrane of umbrella cells.
The antibody of clone AU1 reacts specifically with Uroplakin III. In formalin-fixed paraffin sections Uroplakin III can be detected in 50 – 60 % of primary urothelial carcinomas and metastatic carcinomas of urothelial origin. Uroplakin III is known as specific urothelial differentiation marker in the context of metastatic carcinomas of unknown origin. Benign
Brenner tumours, but not transitional cell carcinomas of the ovary, are the only exceptions from urothelial expression of Uroplakin III described so far.
The immunohistochemical detection of Uroplakin III often results in irregular allotted signals, sometimes only with focal positivity. Additionally, cytoplasmic staining of varying intensity may occur besides the membrane staining.
